I've spent time in and around Elkhardt, Goshen, Harrisonburg, and Hutchinson, but I have actually never been to Lancaster County.
I'm going to be passing through in the near future, without a lot of time. Is there anything in particular I should not miss?

The Hans Herr house, railroad museum of Pennsylvania and Lancaster central market are what this Marylander would suggest.
Lots of tourist traps out on 30 east of town.

These both look really cool. How much time would I need to see them?

A minimum of 2 hours at each place.
